The reaction of cytochrome oxidase with cyanide.

چکیده
The ability of cyanide to inhibit the oxygen consumption of biological systems has long been known (cf. (4, 13)). Warburg (12, 13) appears to be the first to suggest that this action of cyanide was due t,o its combination with the ferric form of the enzyme, called by hi the Atmungaferment and later called cytochrome oxidase by Keilin and Hartree (5). In this paper we make use of this reaction of cyanide with cytochrome oxidase to obtain some quantitative spectroscopic data on the reduced form of this enzyme and to draw certain conclusions concerning the nature of this enzyme whose positive identification has been somewhat elusive.

EFFECT OF CYANIDE IN CARDIOPLEGIC SOLUTION ON ISOLATED RAT HEART FUNCTION
It has been known that cardiomyocytes possess a remarkable ability to downregulate their energy expenditure on restricted 02 supply. However it has also been speculated that an ability to suppress aerobic respiration and ATP utilization could be a protective response to prolonged hypoxia. Having the role of the cytochrome oxidase enzyme in mind, one can ask if its inhibition by using cyani...
متن کاملStudies on cytochrome oxidase. II. A reaction of cyanide with cytochrome oxidase in soluble and particulate forms.
The extent of inhibition of cytochrome oxidase activity by cyanide depends upon the method of assay. It has been contimed that this inhibition is “readily reversible” with the use of the standard manometric technique. However, in the spectrophotometric assay only about 10% of the control activity is observed after the cyanide is removed by physical methods. The polarographic assay yields interm...
متن کاملA spectrophotometric study of the competition of methemoglobin and cytochrome oxidase for cyanide in vitro.
Although the efficacy of induced methemoglobinemia in cyanide poisoning has been demonstrated both therapeutically and prophylactically (l),l the tissue reactions involved in the protection of animals from the lethal effects of the cyanide ion by methemoglobin are only imperfectly understood. Presumably, cyanide interferes with cellular respiration by forming a slightly dissociable complex with...
متن کاملKinetic studies on the binding of cyanide to oxygenated cytochrome c oxidase.
The reaction of cyanide with oxygenated cytochrome c oxidase was followed by means of flow-flash techniques. The oxygenated form, produced after photolysis of the partially reduced CO complex in the presence of cyanide and O2, shows cyanide-binding properties distinct from those of both the oxidized and the reduced forms of the protein. The binding is a single process (k = 22M-1-S-1) linearly d...
متن کاملThe reaction of cytochrome oxidase with cyanide. Preparation of the rapidly reacting form and its conversion to the slowly reacting form.
The magnitude of the slow phase of reaction of cytochrome oxidase with cyanide has been correlated with the size of the epr signal at g' = 12. This epr signal was not found in submitochondrial particles, and significant g' = 12 epr was only observed late in the purification of solubilized enzyme. The Hartzell-Beinert procedure for the purification of cytochrome oxidase (Hartzell, C.R., and Bein...
